Scalp Comfort Follows What Inflammation Does to Skin

Scalp comfort is a skin question before it is a hair-count question. The scalp is skin that houses follicles. When inflammatory mediators rise, skin swells, neutrophils arrive, and, in culture, human follicles stop growing and keratinize abnormally. Folli-Activ includes aloe vera extract for the published anti-inflammatory and skin-trial record of aloe gel. That record is not an oral hair-count trial of this capsule.

What inflammatory signals do to a follicle

Philpott, Sanders, Bowen, and Kealey kept human hair follicles in culture and added cytokines. Interleukin-1 alpha, interleukin-1 beta, and tumour necrosis factor-alpha were potent inhibitors of growth. The histology of inhibited follicles was a dystrophic anagen picture: condensed dermal papilla, vacuolated matrix, abnormal keratinization of the bulb and inner root sheath, and disrupted melanocytes. A scalp that is biochemically inflamed is a scalp whose follicles, on this evidence, are exposed to signals capable of stopping growth and scrambling keratinization. Comfort, in that frame, is not a cosmetic adjective. It is the absence of those signals in the tissue around the follicle.

The Philpott experiment did not test aloe. It defines the problem. The aloe papers define what aloe gel did in inflammation assays and in topical skin trials.

Edema, neutrophils, and prostaglandin E2

Vázquez, Avila, Segura, and Escalante applied aqueous, chloroform, and ethanol extracts of Aloe vera gel to two carrageenan models in rats: paw edema, and neutrophil migration into the peritoneal cavity. Aqueous and chloroform extracts reduced both edema and neutrophil entry. The ethanol extract reduced neutrophils and did not reduce edema. The aqueous extract inhibited prostaglandin E2 production from radiolabelled arachidonic acid, which is a cyclooxygenase readout. Reference drugs indomethacin and dexamethasone reduced edema and neutrophil migration in the same models. The gel, prepared as those extracts, has a measured anti-inflammatory effect on swelling, on neutrophil traffic, and on PGE2.

Bautista-Pérez, Segura-Cobos, and Vázquez-Cruz added bradykinin. Crude gel fraction F-1 cut the maximal bradykinin contraction of rat ileum by 10 percent. A salt-precipitated fraction cut it by 22 percent. A column fraction cut it by 60 percent. The active subfraction F-GB cut it by 67 percent. Bradykinin is a separate inflammatory peptide from the arachidonic acid path. Aloe gel contained material that inhibited it, and the activity concentrated into one fraction. That concentration step matters: activity was a property of a component of the gel, tracked through purification, not a story told about the whole leaf in the abstract only.

Twenty-three controlled trials on skin

Hekmatpou, Mehrabi, Rahzani, and Aminiyan gathered clinical trials from 1990 to 2016 that used Aloe vera gel, cream, or derivatives with a control group. Twenty-three trials met the bar. The conditions in that set were prevention of skin ulcers and treatment of burns, postoperative wounds, cracked nipples, genital herpes, psoriasis, and chronic wounds including pressure ulcers. These are topical uses on skin. Psoriasis and ulcer prevention are the closest neighbours to a comfort conversation, because both are about inflamed or broken skin rather than about a hair count. The review does not include a vertex hair-count protocol, and this page does not add one.

Royal jelly's fatty acid has a parallel skin-barrier dataset, covered in its own essay: synthetic 10-HDA raised filaggrin and hydration in UV-dried skin. Aloe and royal jelly are both in Folli-Activ because both have skin papers. They are not duplicates. Aloe's papers are edema, PGE2, bradykinin, and wound and psoriasis trials. Oral formula, topical evidence, stated as such

Folli-Activ is an oral capsule. The Vázquez experiments were extracts in rats. The Hekmatpou trials were gels and creams on skin. The honest product statement is that the oral formula includes aloe vera extract, and that the comfort rationale is this skin and inflammation literature. It is not a statement that swallowing the capsule reproduced a burn-healing trial.
